Assessing the role of Fc?RIIIa expression on CD8 T cells

Fc? receptors (Fc?Rs) are ubiquitously expressed by nearly all lineages of innate and adaptive leukocyte populations, with the notable exception of T cells, which are negative for all classes of Fc?Rs. While at steady state T cells are negative for all Fc?R classes, we observed that a subset of CD8 T cells specifically induces the expression of the activating Fc?R, Fc?RIIIa, in response to viral infection. We used scRNAseq to compare the transcriptional profile of Fc?RIIIa+ and Fc?RIIIa- CD8 T cell populations from the lungs of influenza-infected Fc?R humanized mice. Overall design: Cells (NK cells and T cells) from the lungs of influenza-infected Fc?R humanized mice were isolated by FACS and analyzed by scRNAseq to determine the gene expression profile and T-cell receptor repertoire.
