The goal of this study was to determine the gene expression pattern of the Saccharomyces cerevisiae model (cbf5-D95A) of X-linked Dyskeratosis Congenita. We conducted high-resolution sequencing of the transcriptomes from cells carrying CBF5 and cbf5-D95A using an RNA-seq approach. An array of computational tools was applied to map reads to the S. cerevisiae genome. mRNA abundance was determined for the yeast genes, helping to explain the stress caused on the yeast cells carrying the cbf5-D95A mutation. A second experiment with WT, cbf5-D95A, upf1?, and cbf5-D95A upf1? was done in an effort to identify messages that had lower abundance in cbf5-D95A cells and were rescued in upf1? cells, and which contained predicted -1 PRF signals.
