In order to explore and compare the transcriptomic profile of leucocytes and stromal cells present in the dermis and hypodermis of C57BL/6 mice, isolated cells from each of these skin layers were subjected to single-cell RNA-sequencing analysis. Back skins from 3 adult C57BL/6 female mice were harvested and the dermal and hypodermal layers isolated by mechanical force and enzymatic treatments. Cells were liberated from each skin layers after enzymatic digestion of the tissues. Leukocytes (CD45+) and stromal cells (CD45-) cells were isolated by fluorescence-activated cell sorting (FACS) and their transcriptome assessed by single-cell RNA-sequencing using the 10X genomics plateform.
