Glucocorticoid recptor knockdown in primary amnion epithelial cells treated with MPA and TNFa

To analyze the effect of glucocorticoids on primary amnion epthelial cells, we harvested tissue samples from 10 healthy ceseaean section patients. We knocked down the glucocorticoid receptor with an siRNA or used a control siRNA. We treated samples with medroxyprogesterone acetate. Half the cells we subjected to a TNFa challenge Primary amnion epithelial cells were transfected with a glucocorticoid receptor knockdown or control siRNA and then 72 hours later treated with 10-6M MPA for 6 hours . A subset of cells were them treated with a TNFa challenge for 24 hours.
