In vitro integration of CRISPR spacers - Streptococcus thermophilus

To better understand the mechanism and requirements for integration of new spacers into CRISPR arrays, we carried out in vitro integration experiments. Cas1 and Cas2 from a type II-A systems (Streptococcus thermophilus) were incubated with pre-spacer and DNA bearing a CRISPR array; the resulting integration events were analyzed to understand how features of the CRISPR repeat guide integration.
