Mass Spectrometry of human cGAS acetylation

HEK293T cells transfected with Flag-cGAS were treated with 10 mM acetate. The cells were then harvested and subjected to immunoaffinity purification using an anti-Flag antibody, with IgG serving as a control. The immunoprecipitated proteins were separated by SDS–PAGE, and the gel was lightly stained with Coomassie Brilliant Blue. The band of interest was excised, digested with trypsin, and analyzed by LC–MS/MS on a Q Exactive Plus mass spectrometer (Thermo Fisher Scientific) coupled online to a UPLC system. The acquired spectra were searched against the human RefSeq protein database using Proteome Discoverer to identify acetylated cGAS peptides.
