NCT05110495: Windows Trial of INsulin-like Growth Factor Neutralising Antibody Xentuzumab in MEN Scheduled for Radical Prostatectomy (WINGMEN)This phase 0 window of opportunity trial offered surgical patients with prostate cancer additional once weekly intravenous (i.v.) xentuzumab infusions (1000mg), during the 4-6-week window they normally wait for prostatectomy. The study IMP is xentuzumab, a humanised IgG1 monoclonal antibody that selectively neutralises the IGF ligands (IGF-1 and IGF-2) to inhibit activation of IGF-1R and INSR-A.Peripheral blood was sampled before the first i.v. infusion with xentuzumab to evaluate pre-treatment levels of peripheral blood mononuclear cells (PBMCs), referred herein as “pre”. This was used to compare with PBMCs collected after treatment with xentuzumab, prior to their surgery, referred to as "post". Samples are paired; i.e. the sample PreWN01 corresponds to the pre-treatment sample, while PostWN01 refers to the post-treatment sample from the same patient.As to investigate potential immune correlates and biomarkers of response to xentuzumab treatment, mass cytometry (CyTOF) was used for deep interrogation of 37 markers and immunophenotyping of PBMCs before, and after treatment with xentuzumab. Hence, this public dataset includes 54 downloadable .fcs files representative of 27 paired patient samples analysed by mass cytometry.
