Few methods exist to accurately identify treatment response in patients with high-grade gliomas. This study demonstrates the potential of using cell-free DNA to monitor relative disease burden over time in patients with high-grade gliomas (HGGs) from plasma samples. By identifying and quantifying tumor-specific amplified DNA junctions in plasma, this approach offers a non-invasive, highly sensitive method for real-time monitoring. The ability to detect high-copy number junctions in plasma correlated with and in some cases preceded changes in radiographic disease burden. This approach may enable rapid analysis of treatment efficacy and early detection of progression once evaluated across a larger cohort of patients.
