We use in vitro selection to isolate, from a random-sequence DNA library, DNA molecules capable of adopting structural arrangements that favour intramolecular circularization over intermolecular ligation by T4 DNA ligase. This approach has led to the isolation of Dsub1 that allows for the large-scale synthesis of DNA rings in both cis and trans reactions with extremely high yield and selectivity.
