Fcγ receptors (FcγRs) are ubiquitously expressed by nearly all lineages of innate and adaptive leukocyte populations, with the notable exception of T cells, which are negative for all classes of FcγRs. While at steady state T cells are negative for all FcγR classes, we observed that a subset of CD8 T cells specifically induces the expression of the activating FcγR, FcγRIIIa, in response to viral infection. We used scRNAseq to compare the transcriptional profile of FcγRIIIa+ and FcγRIIIa- CD8 T cell populations from the lungs of influenza-infected FcγR humanized mice. Cells (NK cells and T cells) from the lungs of influenza-infected FcγR humanized mice were isolated by FACS and analyzed by scRNAseq to determine the gene expression profile and T-cell receptor repertoire.
