In this study, we employed TIER-seq in V. cholerae to identify cleavage sites of RNase E on a genome-wide scale. Overall design: V. cholerae wild-type and rneTS (carrying the rne-3071 allele encoding a temperature-sensitive variant of RNase E) were grown in triplicates at 30°C to late exponential phase, divided in half and grown at 30°C or 44°C for 1h before collection of RNA samples. Following RNA-seq, 5'ends profiles were compared to detect RNase E sites (5' ends missing in the rneTS mutant strain at 44°C compared to the wild-type).
