Dataset Description The dataset, named Selection of DNA Aptamers for Patulin, A Challenging Toxin in Fruit Products for Antibody Production. Contains raw and processed data from selection of DNA aptamers by capture-SELEX, DNA sequencing and analysis, Thioflavin T (ThT) fluorescence titration experiment, and isothermal titration calorimetry (ITC) experiments evaluating the binding interactions between DNA aptamers and a target small molecule patulin. Dataset Composition The dataset comprises 12 sheets in a .CSV file. After sequencing the PCR-amplified DNA products, Geneious-Prime software was used for family classification and conserved sequence analysis. The ThT fluorescence experiment included affinity measurement of aptamers, the effect of different salt ion concentrations on affinity, aptamer selectivity, and fluorescence quantification curves of patulin in fruit juice. ITC experiments include affinity measurements of aptamers and their mutants to targets. Data Collection and Processing Experiments were conducted between May 2024 and May 2025 at the Department of Chemistry, University of Waterloo, using a MicroCal 200-ITC instrument (Malvern Panalytical, UK). The aptamer was dissolved in MES-buffered saline (pH=6.0) and degassed prior to titration. The small molecule was dissolved in MES buffer at the desired concentration. All measurements were performed at 25 °C with an initial reference power of 10 μcal/s. Raw data were processed using Origin 8.0 software (OriginLab, USA) with baseline correction applied to remove instrument drift. Thermodynamic parameters were calculated via non-linear least squares fitting using a single-site binding model. The ThT fluorescence experiment was performed using HORIBA Scientific's FluoroMax-4 test and analyzed using FlourEssence V3.5. They are run at a small percentage ofthe runalong with the 16S amplicon sequencing frothe run's principal customer. We perform qPCR and run your pool on ourAgilent TapeStation to estimate size and theconcentration of your pool to determine theloading amount on the run. Data Format and Accessibility The dataset is provided in CSV format, accompanied by a README.txt file detailing the variable definitions, experimental setup, and data processing pipeline. Ethics and Compliance This study does not involve human or animal subjects. All data were generated in compliance with institutional research guidelines.
