Mammary tumors of parental and aggressive variants of an ErbB2+ breast cancer mouse model that differs in the degree to which is is experiencing oxidative stress as a result of forced expression of an oxidative stress inducer called p66ShcA. NIC are ErbB2+ murine breast cancer cells, which lack p66ShcA. We engineered the parental cells to constitutively overexpress p66ShcA. These NIC/p66Low and NIC/p66High breast cancer cells were serially passaged in the mammary fat pads of mice and after two rounds of in vivo selection, the cell lines were explanted back into culture. Both explanted populations were more aggressive (higher tumor growth rate and increased metastasis) compared to their parental counterparts. RNAseq was performed on bulk tumors (3 tumors each) from the following groups: NIC/p66Low-Parental; NIC/p66Low-Aggressive; NIC/p66High-Parental; NIC/p66High-Aggressive.
