Escherichia coli strain 53638. This strain (53638; serotype O144) is an enteroinvasive Escherichia coli (EIEC) isolate that was obtained from Maj. Carl Brinkley at the Walter Reed Army Institute of Research. Based on phenotypic and biochemical evidence the strain is more similar to E. coli than Shigella and represents an intermediate point between them. This strain is positive for IpaB, a protein that is involved in vacuole lysis and intracellular survival in Shigella, and the infection process occurs in a similar manner, with an intracellular stage of replication followed by cell-to-cell spreading. Pathogenicity has been confirmed in human challenge experiments. Two plasmids typically found in Shigella, pINV and pCRY, are also found in this strain. Additional features include the serotype O144, the inability to utilize lactors, and the lack of toxin production. The stock used for genomic DNA preparation was obtained from a master- and production cGMP cell bank available at WRAIR/NMRC. Two libraries were constructed - a small insert library (2-3 kb) and a large insert library (10-12 kb) from which 11,853 and 52,441 reads were sequenced, respectively.
