The nuclear envelope transmembrane proteins (NETs) NET29a/Tmem120a and NET29b/Tmem120b are expressed or localise preferentially to the nuclear envelope in adipocytes. We knocked these proteins down using specific shRNAs and studied their effect in the diffentiation of the mouse 3T3L1 preadipocyte cell line. Total RNA was isolated from 3T3L1 cells at d0 (confluent preadipocytes) and d6 post induction of differentiation using medium supplemented with 0.5uM insulin, 1uM dexamethasone, 1uM rosiglitazone and 0.5mM IBMX, replacing the medium every 48h up to 9 days post-induction with medium supplemented with insulin and rosiglitazone only. Tmem120a/b knock-downs were generated by transduction of shRNA-encoding lentiviruses as described in Batrakou et al. (2015) PLoS ONE 10(05): e0127712.
