This study utilizes the robotic OT-2 platform to systematically investigate the effects of varying pipetting speeds on yeast growth and gene expression. By employing RNA-seq and growth assays, we observed no significant differences in yeast growth and gene expression within the range of pipetting speeds investigated. Our findings reveal the potential of robotic platforms for optimizing culture conditions and enhancing reproducibility and accuracy in biological experiments, thus providing valuable insights for future research. Overall design: To investigate the effect of four different pipetting speeds controled by liquid handling robot on gene expression and growth speed, we performed RNA sampling and growth assays using liquid handling robot with four different pipetting speeds, two biological replicates, and three technical replicates. We then performed gene expression clusttering analysis using data obrained from RNA-seq of 24 samples.
