The goal of this study was to define the impact of beta-glucan on the liver This repository contains single cell RNA sequencing of mouse livers with intra-portal injection (8572-YFP mouse tumor cells) (n=3 samples/condition) untreated or 14 days post-treatment with beta-glucan x3. Tissues were digested in RPMI media containing collagenase/DNAse and passed through a 40 μm strainer. Subsequently, dead cells were removed using the Dead Cell Removal Kit (STEMCELL Technologies). CD45+ cells were isolated using the CD45+ isolation kit (STEMCELL Technologies). Kupffer cells were enriched by layering the flow through from the CD45+ selection step over Ficoll Paque Plus and cells from the buffy coat combined with CD45+ cells for sequencing. Single-cell cDNA libraries were prepared at the CHOP Single Cell Genomics Core at the University of Pennsylvania.
