We used bulk RNA-seq to compare gene expression profiles of nitrergic (NO) neurons vs other neurons in control hPSC-derived ENS cultures vs cultures that were enriched in NO nuerons by treating their progenitors with PP121. Using H9 NOS1::GFP resporter hPSC cells, we generated enteric nervous system (ENS) cultures treated with or without PP121 which was identified to enriched nitrergic (NO, NOS1+) neruons. We then sorted the neruons (CD24+ cells) into NO (GFP+) vs other (GFP-) and performed bulk RNA-sequencing.
