RT2 Profiler PCR Array Gene Expression Analysis Report

Total RNA was isolated from ldSCAPs and hdSCAPs cultured in 75 cm² flasks at passage 7 using the RNeasy Mini Kit (QIAGEN, USA), according to the manufacturer’s instructions. Complementary DNA (cDNA) was synthesized using the RT² First Strand Kit (QIAGEN).

Gene expression profiling was performed using the QIAGEN Human EMT RT² Profiler PCR Array (Cat. No. PAHS-090Z), which quantifies the expression of 84 genes associated with epithelial–mesenchymal and mesenchymal–epithelial phenotypes. Real-time PCR was performed according to the manufacturer’s protocol. The array includes mesenchymal-associated markers (e.g., VIM, CDH2, collagens), epithelial-associated markers (e.g., CDH1 and cytokeratins), EMT-related transcription factors (e.g., SNAI1, SNAI2, TWIST1, ZEB1, ZEB2), and signaling molecules involved in pathways such as TGF-β, Wnt, and Notch.
