ssDNA strands of the form ACAC NNNNNNNN ACAC are exposed to various doses of 266nm radiation up to dosages of 200 absorbed photons per base. The resulting samples are prepared for sequencing using commercial sequencing kits (Swift Accel-NGS 1S) and read out via Hi-Seq (Illumina). Data is used to infer sequence-dependent UV damage yields in high throughput and across a wide range of doses.
