Metagenomic Study of Granisetron Intervention in LPS-Induced Septic Mice

Granisetron (GA), a microbial metabolite, is previously reported to be relative with liver protection under sepsis. However, whether GA is effective in gut protection and how it affects the host- microbiota interactions, are still unclear. Here, we aim to investigate the role of GA in modifying the gut microbiota.Wild-type male C57BL/6 mice (6-8 weeks old) were obtained from the Department of Laboratory Animal Science, Peking University Health Science Center. Following body weight measurement and numbering, the mice were randomly allocated into three groups (n=6/group): Control (Ctr), LPS, and LPS+GA. Based on previous granisetron-related studies, the LPS+GA group received intraperitoneal (i.p.) injections of granisetron (1 mg/kg) 1 h before and 4 h after LPS administration. The LPS group was administered an equivalent volume of PBS at matched timepoints, while the Ctr group received PBS injections at three corresponding timepoints to minimize experimental variability.we collected fecal samples from mice in different groups for metagenomic sequencing to observe the effects of various interventions on the gut microbiome.
