Aim of the study: Quantitative and preliminary qualitative analysis of exosomes (sEVs), the population of small extracellular vesicles secreted in control and hyperglycemic conditions. Methods: Human beta-pancreatic 1.1.B4 cell line was used. Experiments were performed in control (NG) and hyperglycemic (HG) conditions, respectively 5 and 25 mM of D(+)-glucose. Cells were cultured in parallel, each condition consisting of 4 biological replicates. Extracellular vesicles enumeration and size distribution was measured by nanoparticle tracking analysis (NTA), Attenuated Total Reflectance Infrared Spectroscopy (ATR-IR) and Time-of-flight secondary ion mass spectrometry (ToF-SIMS) were used for chemical characterisation, and cryogenic transmission electron microscopy (Cryo-TEM) was applied for size and shape visualisation. Following parameters were measured: saturated to unsaturated fatty acids (SFA/UFA) ratio, lipid to protein ratio, acyl chain length (ACL). The dataset contains raw data belonging to the experiment with respective files with analysis.
