Disturbance of the gut microbiota has been associated with type 1 diabetes (T1D), but the effect of gut dysbiosis on cognitive function in T1D and its potential mechanism remain unknown. In this study, we treated streptozotocin-induced diabetic mice with vancomycin (Vanc) to disturb the gut microbiota and metabolic phenotype. In order to examine the gut microbial changes, we used a 16S rRNA gene sequencing approach to analyze the gut microbiome in caecum contents of normal control (CON), type 1 diabetic (T1D), and vancomycin-treated type 1 diabetic (T1DV) mice at 3, 7 and 11 weeks. To further determine the effect of antibiotic exposure on the gut microbiota at the species level, we performed a metagenomic analysis of the bacteria in caecum contents. Additionally, to study antibiotic effect on host metabolism, a 1H NMR-based metabolomics approach was applied to analyze metabolic changes in caecum contents, feces, caecum tissues, colonic tissues, serum, and hippocampus. To establish a causal link between the gut microbiota and host metabolism, we next transferred fecal material from healthy donor mice (COND) to T1DV recipients for 14 days.
