Hippocampal transcriptomic response of the cancer treatment Tamoxifen in mice

Tamoxifen is a relatively safe cancer treatment for estrogen receptor-positive (ER+) breast cancer. However, the effect of tamoxifen on cognition is not well studied. We used a mouse model of chronic tamoxifen exposure to examine the effects of tamoxifen on the brain, and report behavior and transcriptomic changes. Female C57BL/6 mice were exposed to tamoxifen (n=5) or vehicle control (n=5) for six weeks. RNA sequencing analysis was performed on the whole hippocampi. Briefly, high-quality total RNA was extracted from the hippocampus (RIN > 9.0) at the end of the treatment. Extracted RNA were processed for polyA library preparation strategy followed by RNA sequencing on Illumina Hiseq instrument using 2x150bp Paired-End (PE) configuration. There are n=5 control and n=4 tamoxifen treated (TAM) libraries in this Bioproject. Hierarchical clustering using Pearson correlation matrix analysis identified one of the TAM samples as an outlier (potential library contamination), and therefore it is excluded from further downstream analysis. Sequencing data of the excluded biological sample (Tamoxifen_2, TAM 2) is not provided.
