The in vitro high-throughput human pri-miRNA processing assays were conducted to check whether mismatches and wobble base pairs in the upper stem of pri-miRNAs affects the DROSHA cleavage. Overall design: 151,248 variants generated from three human pri-miRNAs (pri-mir-106a, pri-mir-342 and pri-mir-514a-1) were processed by mutant DROSHA. The original substrates and cleaved products were cloned and sequenced.
