Porcine Astrovirus CRISPR screen in PK15 cells

Although porcine astroviruses are important pathogens, much of their biology, including their cell surface receptors, remains unclear. We used a genome-wide CRISPR knockout library in PK15 cells to screen for genes essential for astrovirus replication. Cells from the PK15 CRISPR library were infected with porcine astrovirus type 1, and surviving cells were subjected to high-throughput sequencing to identify cells that could not be infected with astrovirus.
