We assessed the role of REDD1 gene in skin inflammation using TPA-induced psoriasiform dermatitis in REDD1 KO mice. We also assessed the anti-inflammatory effect of glucocorticoid fluocinolone acetonide in these animals REDD1 KO and wild type (WT) female in the same F1 C57Bl x 129 genetic background mice were shaved on the back, randomly divided in 3 groups and treated topically with 1). Control (Vehicle control, acetone, 200ul/animal) ; 2). 12-O-tetradecanoyl-acetate (TPA, 4 ug in 200 ul of acetone); or 3) TPA+FA. Some TPA-treated animals were pretreated with glucocorticoid fluocinolone acetonide (FA, 2 ug in 200 ul acetone) one hr before TPA application . Skin was harvested in 24 hrs after TPA application, epidermis was mechanically isolated (by scraping), total RNA was isolated and subjected to RNA-sequencing.
