To investigate the role of EZH2 in transcription regulation in mouse primordial germ cells, we isolated Ctrl and Ezh2 CKO male and female PGCs at E13.5. Our data demonstrates that EZH2-mediated H3K27me3 is required for regulating the expression of meiotic genes in the female germ cells. Furthermore, RNA-seq analysis shows that EZH2 is essential for transcriptional repression of retrotransposons. Expression profiles of Ctrl and Ezh2 CKO male and female mouse primordial germ cells at E13.5.

为探究EZH2在小鼠原始生殖细胞(mouse primordial germ cells)转录调控中的作用,我们于胚胎发育第13.5天(E13.5)分离了对照组(Ctrl)与Ezh2条件性敲除(Ezh2 CKO)的雌雄原始生殖细胞。本研究数据表明,EZH2介导的组蛋白H3第27位赖氨酸三甲基化(H3K27me3)修饰对于雌性生殖细胞中减数分裂相关基因的表达调控必不可少。此外,RNA测序(RNA-seq)分析显示,EZH2对于逆转录转座子的转录抑制至关重要。本数据集包含E13.5天时对照组与Ezh2条件性敲除的雌雄小鼠原始生殖细胞的基因表达谱。
