This study conducted whole genome assembly on diploid plant Iris domestica (estimated genome size of 4.35 Gb).Using a PacBio HiFi (61 ×) combined with DNBSeq (78 ×) and Hi-C (145 ×) sequencing strategy, a contig N50 of 6.53 Mb was initially assembled by Hifiasm. Then, 99.34% of the sequence was mounted onto 16 chromosomes using Hi-C technology, resulting in a total assembly length of 4.52 Gb, with scaffold N50 reaching 270.4 Mb, and with BUSCO integrity of 99.05%—indicating high assembly quality.This assembly provides a high-quality reference genome for studies on the phylogenetic evolution of plants in the Iris genus and research on isoflavonoid biosynthesis.
