This experiment is assocaited with a publication defining a tumor supressor role of FBXO11 in the transformation of MDS. We observe decreased expression of FBXO11 is associated with transformation to secondary AML. This experiment was designed to determine the genes regulated by FBXO11. We compared 4 cell populations. 1. MDS-Cas9 (parent MDS-L cells expressing Cas9 - effectively a control), 2. MDSL-Cas9-FBXO11KO (MDS-L cells with Cas9 and FBXO11 sgRNA - effectively FBXO11 knockout), 3. MDSL-Cas9-FBXO11KOv1OE (FBXO11 knockout cells with overexpression of FBXO11 isoform 1 cDNA), and 4. MDSL-Cas9-FBXO11KOv4OE (FBXO11 knockout cells with overexpression of FBXO11 isoform 4 cDNA)
