Obesity with metabolic dysfunction is a significant risk factor for
progression to hepatic steatosis and liver cancer. PGAM5 is a protein
phosphatase that is associated with regulation of mitochondrial
homeostasis and metabolism. Upregulated expression of PGAM5 in liver
cancer correlates with reduced survival in human patients. We
compared gene expression in human hepatocellular adenoma and carcinoma
cell lines (HepG2, Huh7) with and without deletion of phosphoglycerate
mutase 5 (PGAM5). We found that knockout of PGAM5 in the Huh7 (denoted as
11) and HepG2 (denoted as G4) cell lines results in a dramatic shift in
transcripts of mediators of lipid metabolism. This dataset includes single
cell RNA sequencing data and lipidomic data from cell lines under
different nutrient conditions.
