Signaling induced biophysical disruption of repressed chromatin domains drives immune cell fate

Cleavage Under Target & Tagmentation samples using anti-H3S28ph, anti-H3.3S31ph, anti-H3K27me3, anti-H3K4me3 and Rabbit-IgG in total germinal center cells (for H3K27me3, resting (0h), stimulated (18, 36 and 60h). For histone phosphorylation resting centroblasts (CB) and stimulated centrocytes (CC) were used. For H3K4me3 stimulated Naive B cells were used. For K27me3 Germinal center B cells were used. Stimulation conditions were as follows: 1, 18, 36 or 60 hours stimulation with 10 µg/ml anti-CD40 (Thermo Fisher Scientific 16-0402-86), 10 µg/ml anti-IgG (Thermo Fisher Scientific 16-5098-85), 10 µg/ml anti-IgM (Jackson ImmunoResearch 115-005-020) and 5 ng/ml IL-21 (R&D Systems 594-ML-010).
