The objective of this experiment was to use Nanopore native RNA sequencing (dRNA) with Schizosaccharomyces pombe during vegetative growth in a rich custom medium to uncover the full range of polyadenylated transcripts in the species at very high coverage. With dRNA transcripts, we identified novel alternative isoforms that affect the coding sequences of several coding genes and more than 200 novel transcripts that were not in the annotations. Furthermore, as we use dRNA data we were able to measure poly(A) tails in our transcripts and relate them with their gene expression.
