We report a novel method to quantify bulk changes of chromatin associated proteins in different cellular states and apply it to the mammalian cell cycle Overall design: Sequencing of DNA from Cesium Chloride fractionated chromatin. This purification has been traditionally done to clean-up chromatin before ChIP, but hasn't been sequenced before. The proteins in this chromatin fraction were used for mass spec analysis, with the goal of quantifying bulk chromatin-protein changes in different cell states. We performed high throughput sequencing to determine whether purification of chromatin in this manner led to biases in which genomic regions we were analyzing. This series also contains ATAC-seq data for T98G cells synchronized in G1 and Mphase of the mammalian cell cycle.
