Triple-negative breast cancer (TNBC) bone metastasis has been lacking targeted therapy options. Our research has identified the androgen receptor (AR) as a potential driver of TNBC bone metastasis. To investigate the role of AR in TNBC bone metastasis, we utilized the iliac artery injection method to establish mouse models of bone metastasis. Subsequently, we administered PBS/DHT injections to explore the effects of AR on TNBC bone metastasis in mice. Overall design: BALB/c Nude mice were subjected to iliac artery injection of MDA-MB-231 tumor cells. Subsequently, every three days, intraperitoneal injections of PBS/DHT were administered. After three weeks, bone metastatic tumor tissues were harvested and lysed to obtain single-cell suspensions for single-cell sequencing analysis.
