To further distinguish the heterogeneity among HSCs of young and aged mice, HSPCs (LSK, LinlowScal+c-Kit+) were isolated using a previously validated immunophenotyping strategy . The single-cell gene expression profiles were generated using a customized panel of HSC maker genes, which were identified according to previously published literature . C57BL/6J mice were purchased from JAX (Bar Harbor, ME, USA) and maintained under Specific-pathogen-free (SPF) conditions for aging. All animal studies were performed in accordance with the guidelines approved by the Institutional Animal Care and Use Committees of Sichuan University. HSCs (LinlowSca1+c-Kit+) isolated from young (12wks) and old mice (18months) for single cell RNA sequencing.
