we investigated the functional relevance of these two complexes by deleting AMT6 or AMT7 to generate knockout cells, respectively. To directly interrogate roles of AMT6 and AMT7 in 6mA regulation, we performed Single Molecule, Real-Time (SMRT) sequencing with the Circular Consensus Sequences (CCS) mode of vegetative genomic DNA, AMT6KO and AMT7KO cells of the Tetrahymena MAC genome.
