DNA methylation alteration marks phthlate mediated transgenerational inheritance in murine allergic diseases. Pregnant women attending E-Da Hospital for routine prenatal examinations in their first trimesters were consecutively recruited. We excluded pregnant women: 1) who had history of systematic diseases such as cancer, hypertension and diabetes, chronic use of corticosteroids or immunosuppressant drugs; 2) whose age was older than 45 years; and 3) whose pregnancy was multiparous. Then, the eligible study women were interviewed by a standardized questionnaire and blood and one-spot urine specimens were collected. The same study protocol was also applied to their second and third trimesters, during delivery, and 2-3 weeks after delivery, except the questionnaire was changed to one short diet questionnaire. During delivery, additional specimens of maternal stool, amniotic fluid, placenta, cord blood, and baby meconium were also collected and, 2-3 weeks after delivery, another additional specimen of breast milk was collected. All these different biospecimens have been aliquoted and stored frozen at -70oC for different studies of gene-environmental effect on pregnant women and their offspring. Human placenta samples were collected and clasified in two groups including low and high DEHP exposure. Each group contains 6 samples. All chosen samples were done Bisulfite Epic Illumina microarray.
