Identification of the R-loop regions in the moss Physcomittella patens

R-loops are three-strand nucleic acid structures, consisting of a DNA-RNA hybrid and a displaced single-strand DNA, and form during transcription and conflicts between transcription and replication. Physcomitrella leaf cells arrest at the late S phase and reenter the cell cycle after wounding. Based on our experiment results, we assume that the R-loop formation contributes to the unusual cell cycle arrest and loss of R-loop leads to the cell cycle reactivation. To test this hypothesis, we focus on identification of the R-loop region in leaf cells.
