MicroRNAs are likely to play pivotal roles in both stem cells and cancer, and are further promising candidates for future therapeutic purposes. To understand the role of miR206 in mammary cell differentiation, we profiled the transcription of mamary stem-like cells, comparing control HC11 cells (unspecific miRNA mimic treated) with HC11 cells transfected with miRNA-206 mimic. Overall design: Comparison of NC mimic-treated vs.miR-206 mimic treated HC11 cells. Mouse Operon’s long oligonucleotide- spotted arrays were used (Microarray Inc.,Huntsville, Alabama). Microarray analysis was based on hybridization between the transcript and its corresponding probes. 2-color comparative microarray assays were carried out that each sample (Control and miRNA206 treatment) was labeled with either Cy3 or Cy5 dye. Errors introduced by differential labeling were adjusted using dye-swap replicates.
