This study investigates the effects of olaparib, a PARP inhibitor, and its combined administration with cisplatin on the gut microbiota of female ICR mice. Mice were randomly divided into four groups: control, cisplatin-only, olaparib-only, and a cisplatin-olaparib combination group. Fecal samples were collected at 1, 3, and 6 months after drug administration to assess changes in microbiota over time.DNA was extracted from mouse stool samples using the ISOSPIN Fecal DNA Kit. The V3-V4 region of the 16S rDNA gene was amplified using specific primers. Following amplification, adapter and index sequences were added as part of the 16S Metagenomics Sequencing Library Preparation protocol. The prepared libraries were sequenced using paired-end sequencing on an Illumina platform to assess changes in the gut microbiota.
