Bexarotene and carvedilol combination treatment prevents the EMT process in HMEC-hTert cells through the actions of ARID1A

Previously we found that the combination of the RXR selective agonist, bexarotene (Bex), and the non-selective beta-blocker, carvedilol (Carv), showed synergistic anti-tumor effects in the formation of Her-2 induced breast cancers. In addition, Carv enhanced the anti-proliferative effects of Bex in HMEC-hTert cells. ARID1A is a chromatin remodelling factor that was up-regulated selectively by Bex and Carv.In this study, we found ARID1A genomic occupancy to regulatory elements assigned to genes involved in the transforming growth factor Beta signaling pathway is enriched under Bex+Carv exposure in HMEC-hTert cells. ARID1A and BRG1; ATPase subunit of the SWI/SNF complex, enrichment to their target regions was associated with a change in the expression of target genes, encompassing the epithelial-mesenchymal transition process.
