Background: Ulcerative colitis (UC) is considered an important health problem due to its complex pathogenic factors, which are often accompanied by intestinal environmental disorders and mucosal barrier damage. Oral administration is considered the most promising approach for UC therapy. In order to solve the problems of limited biological activity and short residence time caused by conventional oral administration, an oral nanoparticle PHPP-Eud with a PH-responsive outer membrane was prepared in this study to achieve targeted delivery of traditional Chinese medicine active substances into the colon.Result: In vitro and in vivo studies have shown that PHPP-Eud reduces inflammation, repairs the intestinal mucosal barrier, and increases the proportion of beneficial bacteria, including Lactobacillus and Bifidobacterium, for the treatment of ulcerative colitis. In addition, through the study of the mechanism of PHPP-Eud, FoxO4 is expected to become an important target for the treatment of ulcerative colitis.Conclusion: PHPP-Eud can be used as a PH-responsive nanoparticle for the treatment of ulcerative colitis, and new therapeutic targets have been developed for its treatment.
