To elucidate the role of DOCK7 in melanoma progression, we conducted both in vitro and in vivo functional assays to investigate DOCK7's function in cell proliferation, colony formation, and invasion. Gene expression was carried out the validate the knockdown efficiency before continuing the subsequent experiments. We also performed mass spectrometry and RNA sequencing analysis for better characterization of the molecular role of DOCK7 in melanoma development. HIF1a and ADAM10 was identified from mass spectrometry and RNA sequencing. The in vitro invasion assay conducted on knockdown of HIF1a and ADAM10 showed remarkable reduction in invasiveness of melanoma cells, underscoring their potential to work with DOCK7 in promoting melanoma invasion.
