该数据集包括了2021年至2024年,从青藏高原玉珠峰、达索普冰川、枪勇冰川冰缘带、昂仁金错、崩错、邦达错、打加错、达瓦错、达则措、扎当冰川、宁金岗桑冰川、东绒布冰川、芦布错、宁金岗桑、纳木错、普莫雍错、错鄂、玉珠峰冰川、拉昂错、戈芒错、宗雄错、松木希错、结则茶卡、龙木错、吴如错、巴木错、芒错、蓬错、纳木措、达热布错、恰规错、张乃错、昂古错、美玛错、佩估错、东绒布、齐格错、木吉冰川、羊卓雍错、左秋普、拉果错、雅拉冰川、老虎沟冰川、枪勇冰川、杰马央宗冰川、廓琼岗日冰川、古里雅冰川、唐古拉冰川、老虎沟、木嘎岗琼冰川、拉萨、山南等地采集的不同生境(冰川冰雪、冰芯、冰尘、土壤、河流、湖泊、湖泊沉积物以及人居环境)样品中分离得到的特殊微生物信息。以excel表格的形式体现,其中包括物种名称、基因序列号、菌种保藏时间、菌株分离所用的培养基类型、最适培养温度、菌株颜色以及菌株来源等信息。表格中”-”表示没有该信息。这些特殊微生物表现出不同的菌落颜色,这可能与微生物适应青藏高原强辐射、低温、寡营养的环境有关,通过产生不同的色素等来适应严酷的环境或者来实现抗生素抗性、耐药性等功能。这些微生物是通过稀释平板涂布,在不同温度下培养(培养基为R2A等)分离,确定无其他杂菌污染、为单菌落之后获得。得到的单菌落后续会进行16S rRNA全长测序,通过97%的相似度确定其是否为新的种;如果该序列与最相似菌株的序列相似度低于97%,会进行全基因组测区,确定该菌株在相应的属、科甚至门类中的系统发育关系,确定该菌株是否具有新的系统发生地位,增加了青藏高原微生物多样性,拓宽了对具有抗生素抗性等特殊功能微生物的认知。
This dataset contains information of specialized microorganisms isolated from environmental samples collected across various sites on the Qinghai-Tibet Plateau between 2021 and 2024, including Yuzhu Peak, Dasuopu Glacier, Qiangyong Glacier periglacial zone, Angren Jinco, Bengco, Bangdaco, Dajiacuo, Dawacuo, Daze Co, Zhadang Glacier, Ningjin Kangshar Glacier, Dong Rongbu Glacier, Lubu Co, Ningjin Kangshar, Namtso, Puma Yumco, Co Ngo, Yuzhu Peak Glacier, La'ang Co, Gomang Co, Zongxiong Co, Songmu Xi Co, Jiezha Caka, Longmu Co, Wuru Co, Bamu Co, Mang Co, Peng Co, Dahuobu Co, Qiaqui Co, Zhangnai Co, Anggu Co, Meima Co, Peiku Co, Dong Rongbu, Qige Co, Muji Glacier, Yamdrok Lake, Zuoqiupu, Laguo Co, Yala Glacier, Laohugou Glacier, Qiangyong Glacier, Jiemayangzong Glacier, Kuo Qiong Gangri Glacier, Guliya Glacier, Tanggula Glacier, Laohugou, Mugagangqiong Glacier, Lhasa, and Shannan. The sampled habitats cover glacial ice and snow, ice cores, cryoconite, soil, rivers, lakes, lacustrine sediments, and human-inhabited environments.
The dataset is provided in an Excel spreadsheet, which includes details such as species names, gene accession numbers, strain preservation dates, media types used for strain isolation, optimal growth temperatures, colony colors, and strain sources. The symbol "-" in the spreadsheet indicates missing corresponding information.
These specialized microorganisms display diverse colony colors, which is likely associated with their adaptation to the harsh Qinghai-Tibet Plateau environments characterized by intense ultraviolet radiation, low temperatures, and oligotrophic conditions. They may produce various pigments to tolerate the extreme habitats, or develop functions including antibiotic resistance and drug resistance.
These strains were isolated via the dilution plate spreading method, cultured at different temperatures using media such as R2A, and purified to obtain axenic single colonies free of exogenous microbial contaminants.
Subsequently, full-length 16S rRNA sequencing was performed on the isolated single colonies. Whether a strain represents a novel species is determined based on a 97% sequence similarity threshold: if the sequence similarity between the isolate and its closest matched strain is below 97%, whole-genome sequencing will be conducted to clarify the phylogenetic relationship of the strain within its corresponding genus, family, or even phylum, and verify whether the strain has a novel phylogenetic status. This dataset enriches the microbial diversity inventory of the Qinghai-Tibet Plateau and broadens the scientific understanding of microorganisms with special functions such as antibiotic resistance.
