We demonstrated that CELF2 regulates FAT10, and promotes mTORC1 signaling pathway activation in MLL-AF9-AML mice. Comparative gene expression profiling analysis of RNA-seq data for celf2 KO+MA9 mice and MA9 mice. Celf2 conditional deletion mice were generated using the homologous recombination technique to selective knockout of exon 5 which causes in a frameshift mutation, resulting in a stop codon (as shown in the figure below). The mice were then mated with Vav1-Cre transgenic mice expressing Cre recombinase under the control of the Vav1 promoter to delete Celf2 in hematopoietic cells at the embryonic stage. As expected, half of the offspring were Celf2 wild-type, and the other half were Celf2 knockout mice. Continue breeding with Celf2fl/fl mice to obtain Vav1-Cre;Celf2fl/fl mice.
