The aim of this basic research is to test the efficacy of intranasal administration of Galanin-like Peptide (GALP) and its clinical application under the hypothesis that GALP prevents obesity in mice fed a high-fat diet (HFD). Focusing on the mechanism of regulation of lipid metabolism in peripheral tissues via the autonomic nervous system, we confirmed body weight changes after intranasal administration of GALP in diet-induced obesity (DIO) mice. Whole genome DNA microarray was carried out on the liver samples (liver tissue of DIO C57BL/6J mice was ground into a fine powder in liquid nitrogen and stored at -80°C until RNA isolation, and total RNA was extracted from powdered mouse liver tissue using the RNeasy Mini Kit ) to aimed to confirm the anti-obesity effects of intranasal administration of GALP in the diet-induced obesity (DIO) model mice. To detect significantly differentially expressed genes between control (n=8) and GALP-treated (greater weight loss group, n=4) and control and GALP-treated (less weight loss group, N=4) samples, total RNA extracted from mouse liver tissue for each control (n=8) , GALP-treated (greater weight loss group, n=4) and GALP-treated (less weight loss group, N=4) was pooled in each group, prior to DNA microarray analysis.
