CIC binding was determined in KP murine lung cancer cells in untreated cells as well as after treatment with trametinib to enhance CIC binding to DNA. As a negative control, CIC binding was also determined in KPCic cells that express a non-functional CIC incapable of DNA binding. Overall design: Cells were grown in DMEM supplemeted with 10% FBS. Cells were fixed with 1% formaldehyde for 10 min. After quenching and cell lysis, chromatin was sonicated using a Bioruptor Plus Diagenode. Library construction and sequencing was carried out by HaploX Gene Tech Limited (Hong Kong)
