Candida albicans Genome sequencing

We are investigating the genes that are required to grant azole resistance in the clinical isolate strain CaCi-17 isolated by Theodore White. This strain has a gene deletion of a novel resistance determinant that we have identified. We hope to better characterize the mechanisms of resistance of C. albicans by generating resistant isolates, which are then used for subsequent whole genome sequencing. Resistant isolates were generated by plating ~2x10^8 cells on solid YPD or SD laden with high concentrations of azoles (18-26 ug/mL). Resistant isolates were then isolated and validated by microbroth dilution minimum inhibitory concentration assays. Sequence data was generated using an Illumina HiSeq2500 with paired end 125 bp reads. Libraries were prepared using the Nextera XT protocol.
