In order to investigate molecular mechanism underlying Reyan No. 2 BLCs development, transcriptomic analysis was conducted in Reyan No. 2 root tips at 24 and 48 h after germination. For each time course, three libraries were constructed and produced around 50 and 52 million raw reads, and 40.5 and 45.5 million clean reads, respectively. A total of 2036 genes, which were involved in various biological processes, such as the cell wall modification, exhibited differential expression patterns between 48 h and 24 h after germination. Among these differential expression genes (DEGs), 467 genes were up-regulated, while 1569 genes were down-regulated. Furthermore, a total of 90 DEGs were predicted to be involved in cell wall modification, with 16 genes up-regulated and 74 genes down-regulated. The qRT-PCR assay was further conducted to investigate the expression levels of 12 randomly selected DEGs, and results were consistent with the assay of transcriptomic analysis transcriptomic analysis was conducted in Reyan No. 2 root tips at 24, 48 and 72h after germination
